(not a sitcom but still sadness in comedy)

If you ignore the laughter track at the start it gets very poignant at the end

Good call Chimpster, Paul Whitehouse is a genius.

I don't know if there's a clip of this anywhere, and if there is it's only in black and white as for decades it was a "lost" episode which was only found a few years ago, but there's a wonderful moment in the episode of "Dad's Army" called "The Battle of Godfrey's Cottage" when intelligence reports wrongly warn that the German invasion of Britain is under way.

The rest of the platoon have gone off somewhere leavin just Capt Mainwaring, Corporal Jones and Private Frazer. They have only their rifles, a machine gun and a few grenades. Mainwaring looks at the map, and as there's only one road up from the beach (which passes Godfrey's cottage) he decides that that's where they'll make their stand and try and buy as much time as possible for reinforcements to get there.

His usual pomposity is gone, and as he looks at the map he's knows the hopelessness of the situation. He shakes his head, turns to the other two and quietly says "we'll just have to hold them as long as we can.....it'll be the end for us, but I think we're prepared for that" The other two just nod their heads and say "yes, Sir" and these three brave men pick up their weapons and head off expecting that they are going to their deaths in the defence of their country.

I saw a post to scrubs earlier but here is a collection I came across. Scrubs was so well done, I've had tears from both it being really funny and really sad too. I miss it..
